Identification of duplicates of cassava accessions sampled on the North Region of Brazil using microsatellite markers Acta Amazonica
Moura,Elisa Ferreira; Farias Neto,João Tomé de; Sampaio,José Edson; Silva,Diehgo Tuloza da; Ramalho,Girena Fernandes.
Duplicates are common in germplasm banks and their identification is needed to facilitate germplasm bank management and to reduce maintenance costs. The aim of this work was to identify duplicates of cassava from a germplasm bank in Eastern Amazon, which had been previously characterized both morphological and agronomically. In order to be genotyped with 15 microsatellite loci, 36 accessions were selected. These accessions were classified into 13 groups of similar morpho-agronomical characteristics. All loci were polymorphic, and 75 alleles were identified, with an average of five alleles per loci and H E = 0.66. There were determined 34 pairs of genotypes with identical multiloci profiles and the probability of genetic identity was 1.1x10-12 with...

Identification of leaf rust resistance genes in selected Argentinean bread wheat cultivars by gene postulation and molecular markers Electron. J. Biotechnol.
Vanzetti,Leonardo Sebastián; Campos,Pablo; Demichelis,Melina; Lombardo,Lucio Andres; Aurelia,Paola Romina; Vaschetto,Luis María; Bainotti,Carlos Tomás; Helguera,Marcelo.
Leaf rust, caused by Puccinia triticina Eriks. is a common and widespread disease of bread wheat (Triticum aestivum L.), in Argentina. Host resistance is the most economical, effective and ecologically sustainable method of controlling the disease. Gene postulation helps to determine leaf rust resistance genes (Lr genes) that may be present in a large group of wheat germplasm. Additionally presence of Lr genes can be determined using associated molecular markers. The objective of this study was to identify Lr genes that condition leaf rust resistance in 66 wheat cultivars from Argentina. Twenty four differential lines with individual known leaf rust resistance genes were tested with 17 different pathotypes of leaf rust collected from Argentina. Leaf rust...

Identification of Pm24, Pm35 and Pm37 in thirteen Egyptian bread wheat cultivars using SSR markers Ciência e Agrotecnologia
Emara,Hassan Mahmoud; Omar,Ayman Feisal; El-Shamy,Moustafa Mahmoud; Mohamed,Mona Elsaid.
ABSTRACT Powdery mildew of wheat (Triticum spp.) caused by Blumeria graminis f.sp. tritici (DC) E.O. Speer Em. Marchal is one of the most important bread wheat diseases in Egypt. All the Egyptian common bread wheat cultivars are susceptible to that disease at seedling and adult stages. Breeding of resistant cultivars is the most economical and environmentally safe method to eliminate the disease and reduce crop losses. Combinations of two or more effective resistance genes may lead to better, more durable resistance to that disease. Eight Pm genes i.e. Pm2, Pm6, Pm12, Pm16, Pm24, Pm35, Pm36 and Pm37 out of 21 powdery mildew monogenic wheat lines (Pm) were resistant to 42 individual isolates of powdery mildew collected from different governorates in the...

Identification of rapd marker linked to blast resistance gene in a somaclone of rice cultivar Araguaia Trop. Plant Pathol.
ARAÚJO,LEILA G.; PRABHU,ANNE S.; FILIPPI,MARTA C..
The gene Pi-ar confers resistance to Pyricularia grisea race IB-45 in a somaclone derived from immature panicles of the susceptible rice (Oryza sativa) cultivar Araguaia. RAPD technique was used to identify molecular markers linked to this gene utilizing bulked segregant analysis. Initially, the two parental DNAs from the resistant donor SC09 and 'Araguaia' were analyzed using random primers. Of the 240 primers tested, 203 produced amplification products. The two parental DNAs along with the resistant and susceptible bulks of F2 population were screened using 48 primers that differentiated resistant and susceptible parents. Even though eight primers differentiated the resistant bulk from the susceptible bulk, as well as somaclone SC09 and 'Araguaia', only...

Abstract The common bean is characterized by high sensitivity to drought and low productivity. Breeding for drought resistance in this species involves genes of different genetic groups. In this work, we used a SEA 5 x AND 277 cross to map quantitative trait loci associated with drought tolerance in order to assess the factors that determine the magnitude of drought response in common beans. A total of 438 polymorphic markers were used to genotype the F8 mapping population. Phenotyping was done in two greenhouses, one used to simulate drought and the other to simulate irrigated conditions. Fourteen traits associated with drought tolerance were measured to identify the quantitative trait loci (QTLs). The map was constructed with 331 markers that covered all...

Metatranscriptomic analysis of small RNAs present in soybean deep sequencing libraries Genet. Mol. Biol.
Molina,Lorrayne Gomes; Cordenonsi da Fonseca,Guilherme; Morais,Guilherme Loss de; de Oliveira,Luiz Felipe Valter; Carvalho,Joseane Biso de; Kulcheski,Franceli Rodrigues; Margis,Rogerio.
A large number of small RNAs unrelated to the soybean genome were identified after deep sequencing of soybean small RNA libraries. A metatranscriptomic analysis was carried out to identify the origin of these sequences. Comparative analyses of small interference RNAs (siRNAs) present in samples collected in open areas corresponding to soybean field plantations and samples from soybean cultivated in greenhouses under a controlled environment were made. Different pathogenic, symbiotic and free-living organisms were identified from samples of both growth systems. They included viruses, bacteria and different groups of fungi. This approach can be useful not only to identify potentially unknown pathogens and pests, but also to understand the relations that...

Microsatellite markers for identification of a group of italian olive accessions Scientia Agricola
Muzzalupo,Innocenzo; Stefanizzi,Francesca; Salimonti,Amelia; Falabella,Rosanna; Perri,Enzo.
Cultivar characterization for fruit trees certification requires fast, efficient and reliable techniques. Microsatellite markers (SSR) were used in the molecular characterization of 23 genotypes of Olea europaea subsp europaea. The DNA from the olive cultivars was analyzed using nine pre-selected SSR primers (GAPU59, GAPU71A, GAPU71B, GAPU103A, UDO99-01, UDO99-12, UDO99-28 and UDO99-39) and revealed 29 alleles, which allowed each genotype to be identified. In the dendrogram, the nine primers allowed the 23 olive genotypes to be grouped into subgroups corresponding to the same cultivar denominations. SSR markers proved to be efficient and reliable for the molecular characterization of Italian olive cultivars.

Molecular and morphological data from Thoosidae in favour of the creation of a new suborder of Tetractinellida ArchiMer
Luis Carballo, Jose; Bautista-guerrero, Eric; Cardenas, Paco; Cruz-barraza, Jose Antonio; Aguilar-camacho, Jose Maria.
The Thoosidae (Porifera, Demospongiae, Tetractinellida) currently includes the genera Thoosa, Alectona, and Delectona. To this date, molecular data are only available for Alectona. In this study, the phylogenetic affinities of the genera Thoosa and Alectona have been investigated with the species T. mismalolli, T. calpulli, and T. purpurea from the Mexican Pacific using morphology and three molecular loci: the mitochondrial cytochrome oxidase subunit 1 (CO1 mtDNA), 28S rRNA (fragment D2), and 18S rRNA. Morphology and embryology showed that these genera are quite different from the rest of the tetractinellids because larvae of Alectona and Thoosa have unique features in sponges, such as the presence of monaxonic discs in Thoosa and tetraxonic discs in...

Molecular characterization of elephantgrass accessions through RAPD markers Ciência e Agrotecnologia
Passos,Leônidas Paixão; Machado,Marco Antonio; Vidigal,Maria Coletta; Campos,Ana Lúcia.
Elephantgrass pastures are limited by yield variations and reductions in forage quality and availability, thus making the search for genotypes with reduced seasonality a major concern. In order to verify the extent of genetic variability among contrasting cultivars, ten elephantgrass accessions were analyzed through DNA amplification by RAPD technique. A total of 160 DNA bands were generated with the use of 44 random primers and 23% of these bands were monomorphic for all accessions. Gel-obtained binary data (1 for presence and 0 for absence) were used for generating a genetic distance matrix, which was utilized in a UPGMA grouping analysis. Elephantgrass cultivars Cameroon and Vruckwona were the accessions mostly divergent from the others, with an average...

Molecular identification of intergenus crosses involving catfish hybrids: risks for aquaculture production Neotropical Ichthyology
Hashimoto,Diogo T.; Prado,Fernanda D.; Foresti,Fausto; Porto-Foresti,Fábio.
ABSTRACT Monitoring of the interspecific hybrid production and trade is essential for the appropriate management of these animals in fish farms. The identification of catfish hybrids by morphological analysis is unreliable, particularly of juveniles and post-F1 individuals. Therefore, in the present study, we used five molecular markers (four nuclear genes and one mitochondrial gene) to detect hybrids in the trade of pimelodid juvenile fish from different stocks purchased of five seed producers in Brazil. Samples commercialized as pintado (pure species Pseudoplatystoma corruscans ) from three fish farms were genetically identified as hybrid cachapinta (♀ P. reticulatum x ♂ P. corruscans ). In the stocks purchased as cachandiá (hybrid between ♀ P....
